IDT Primer Calculator Calculator Usage Guide

Learn how to use the IDT Primer Calculator calculator and its working principles

How to Use the Calculator

  1. Enter the DNA sequence in the provided field. Ensure the sequence is in uppercase.
  2. Specify the primer length in nucleotides.
  3. Define the desired Tm range (e.g., 18-25°C).
  4. Click the "Calculate" button to compute the melting temperature.
  5. The calculator will display the Tm value and verify if it falls within the specified range.

Working Principle

The melting temperature (Tm) is a critical parameter in primer design. It represents the temperature at which 50% of the DNA duplexes are melted into single strands. The formula used in this calculator is a simplified version that estimates Tm based on the GC content of the primer:

Tm = (G+C) * 2 + (A+T) * 4

This formula assumes a monovalent salt concentration of 50 mM and a DNA stringency of 0.69. Adjustments may be needed for different conditions.
